Russian-born Valentina Tereshkova is credited as being the first woman in space when her ship was launched on 14 June 1963. Incidentally, Sally Ride was the first American woman in space when she went aboard the October 1984 Challenger STS-41G flight.

Everyone knows that women cant drive, especially not space ships. I wouldn't even trust one to park my car, much less land a million-dollar space ship.

The question only asked "in space," not in what capacity. That being said, there were two female Shuttle pilots, both of whom returned safely: Collins (STS-93, 1999) and Melroy (STS-120, 2007).
